Using the profile rewriting automatically prevents most special characters, but it allows spaces.
There's no simple or upgradeable method to limit to a-z0-9 in 4.0.x. Register.php would have to be changed. As a custom project my estimate would be 1 hour.

Hello,
I wanted to say the field "username" in the page of registration of the members,
I would like to prohibit spaces,/,...
(some members uses the address of their Web site "http://www.website.com/" as username).
I want to authorize only the letters of a-z and the numbers of 0-9
